The whole thing reminds me of the Tribes 2 building mod I used to play years back. Nothing I've experienced since has captured that same great feeling you'd get from jumping on a random server and building things with other people in a persistent (as long as the server was up) environment. As is the case with Minecraft, there was virtually no structured gameplay, it was all about the enjoyment of building. Players could even create teleporters and colour coded force fields to keep other players from entering their creations but generally every enjoyed visiting other people's structures and watching each other build.
Remembering how much time I sunk into that small mod makes me realise how something like this can become so popular.

Since you have previous experience with level design tools, I would recommend trying to learn a modeling application such as 3Ds Max / Maya. If that seems too daunting to begin with perhaps try picking up Blender to begin with then work your way up (Blender is free to download).
I think picking up UDK would be a good idea as it's quite easy to import things you have made in the 3D packages into the Level editor. It would also give you some experience with a contemporary video game engine.
I would suggest setting yourself the task of making a simple, small environment. Plan it on paper / in 2D first (read some of the very useful level design tutorials that are available online) then prototype it in UDK. Once that's done try your hand at making some very simple 3D models for it!
Once you've done that you'll be well on your way.

I found the moving menu a little annoying although I liked the roll overs.
Apart from that I like the design and style of the website. Good content as well, especially liked the TF2 train model.

Fun little game to play at work. I really like how slick the interface is, especially the progression overlay on the map at the end.
